EFFECTS OF BAY 11-7082 ON THE FORMATION OF IMMUNOSUPRESSOR DENDRITIC CELLS IN VITRO

Abstract

Dendritic cells (DCs) regulate immune responses by modulating T and B cells in the direction of effector or tolerogenic responses. In this study, the effect of BAY 11-7082 on the formation of immunosuppressive DC of C57Bl/6 mice in vitro using recombinant IL-10 or TGF-β was evaluated. It was shown that the addition of BAY 11-7082 to the culture of immature DCs did not significantly affect the expression of the maturation molecules of dendritic cells (CD80, CD86). When using BAY 11-7082 in combination with IL-10 or TGF-β, reduced the expression of these molecules. The joint cultivation of DC with splenocytes in the presence of BAY 11-7082 and IL-10/TGF-β promoted the differentiation of regulatory T-cells (CD4+CD25hiFoxP3+) and did not contribute to the differentiation of CD4+ T-cells producing IL-10.
