POPULATION NK-CELLS IN PERIPHERAL BLOOD REFLECTS DELINEATED SPECIFIC PROCESSES IN PULMONARY TUBERCULOSIS

Abstract

The purpose of the study was to examine the difference in the number of NK-cells in various forms of pulmonary tuberculosis. During the examined 124 people, of which 99 - have had a form of pulmonary tuberculosis, 25 - were practically healthy people. The study was performed by flow cytometry Coulter Epics XL (BeckmanCoulter, USA). Statistical analysis was performed with the use of programs Statistica for Windows v.6.1. It has been established that the presence of lung tuberculoma is associated with a decrease in the number of NK-cells compared to healthy people by 20 %. Reduced tuberculoma activity is accompanied by lowering of the agreed amount NK-cells by an average factor of two.

Abstract

The purpose of the study is the definition of characteristics of the response of the immune system of patients with lung tuberculoma in the active phase and phase subsides / moderate activity for the establishment of the laboratory criteria for preoperative evaluation of educational activity. The study was performed with the participation of 160 people, We studied the absorption and functional metabolic activity of neutrophils and monocytes, population composition of cells, expression of adhesion molecules and markers of cell activity, the concentration of cytokines. Determine CD11b expression tests and HLA-DR on monocytes, CD11b - on neutrophils, evaluations populations CD3+HLA-DR+ and CD3+CD4+CD25high can be recommended for preoperative assessment of activity tuberculoma lung.

The use of targeted gene sequencing (Roche 454 sequencer) in the patients-representatives of European ethnicity with an excessive amount of strontium in the blood made it possible to detect the presence of a significant number of point replacements in 10 genes: MTHFR - gene of methylenetetrahydrofolate reductase (23 replacements); SULT1A1 - sulfransferase gene (41 replacement); SIRT3 - sirtuin gene (13 replacements); NOS 3 - gene of endothelial NO- synthase (19 replacements), and PPARD - receptor gene (6 replacements), activated by peroxisome proliferator: TERT - telomerase gene (14 replacements); FAS -gene of the death receptor (9 replacements); FOXP3 - transcriptional factor of cellular suppression (4 replacements); TP53 - transcriptional factor of onco suppression (7 replacements); VEGF - gene of vascular endothelial growth factor (8 replacements). Sites of these genes can be proposed as markers to manifest the harm to health caused by chemical factors.

BACTERIA AS PRODUCERS OF CYTOKINE-LIKE SUBSTANCES

Abstract

In experiments in vitro on the 61 clinical isolates S. aureus (n=23), S. haemolyticus (n=10), Enterococcus faecalis and E. faecalis (n=8), Pseudomonas aeruginosa (n=8), enterobacteria - Klebsiella pneumoniae and E. coli (n=12) using a test-system Multiplex (Luminex, USA) to determine the 15 cytokines (G-CSF, GM-CSF, INF-γ, IL-10, IL-12p70, IL-13, IL-17A, IL-φ, IL-2, IL-4, IL-5, IL-6, MCP-1, MIP-φ, TGF-α) studied the production of cytokine-like substances (CLS). Identified and characterized the intergeneric and intraspecific variability of bacteria in their ability to produce CLS in the environment of cultivation. It is established that isolates of S. aureus are the most active producers of CLS. the possible pathogenetic significance of products CLS bacteria and ways to further study this phenomenon were discussed.

Abstract

In experiments in vitro and in vivo it was shown that a synthetic peptide of the active site of GM-CSF -ZP2 (chemical formula: THR NLE NLE ALA SER HIS TYR LYS GLN HIS CYS PRO) stimulates the activity of immune cells (lymphocytes, stem cells, neutrophils, monocytes), inhibits the growth, reproduction and biofilm formation of gram-positive and gram-negative bacteria and has antiviral activity against DNA and RNA viruses and activates the repair of damaged tissues of the organism at the wound process. The presence at this synthetic peptide of such a wide range of immunological effects (immunostimulatory, antibacterial, antiviral, and repair activity) determines the possibility of its use as the basis for creation of new effective medicinal and cosmetic preparations with combined properties that were implemented in the creation of cosmetics ACEGRAM.

FEATURES OF FORMATION OF THE MUCOSAL IMMUNITY OF THE UPPER RESPIRATORY TRACT IN PRETERM INFANTS IN THE FIRST YEAR OF LIFE

Abstract

Studied parameters of local immunity of the mucous membranes of the upper respiratory tract in 47 children the first year of life, born premature. Found that in preterm infants on nasal mucosa there is transient deficiency of IgG, low levels of IgE antibodies, cytokine IF-γ in the second half of life. These features of local immunity of the nasal mucosa is most pronounced in children born at gestational age of 28-32 weeks. The tension of mechanisms of the immune response in the mucosa of the oropharynx shows the excess production of proinflammatory cytokine IL-φ throughout the first year of life, and high IgE and IgA in the first half of the year.

THE INFLAMMATORY PATTERNS IN PATIRNTS WITH BRONCHIAL ASTHMA AND CONCOMITENT OBESITY

Abstract

Currently, it exists a firm tendency of increased prevalence of the excess body mass and obesity all over the world. In serum samples of 108 children and 51 adults with allergic diseases and bronchial asthma of different degree of severity, we measured the levels of ORP, total IgE, leptin, cytokines and IgE Abs against a number of self-tissue proteins by ELISA method. It was revealed the association between body mass index (BMI) and the clinical manifestations of allergy among children, that were characterizes by 2 times higher frequency of excess BMI in asthma group as compared to children with allergic rhinitis (AR) and atopic dermatitis (AD), without BA. It appeared that the elevation of BMI was also associated with worsening of asthma severity degree and the expanding the range of concomitant diseases. Besides of the enhanced levels of total IgE it was estimated the higher frequency of sIgE-response to environmental Ags. In both groups of adults and children, the obesity was associated with increasing of CRP, leptin and TNF-α in serum, being most enhanced in asthma group. The revealed associations confirm the link between obesity, as a chronic inflammatory condition with atopy and development of asthma.

We examined 266 patients 15-17 years old with menstrual disorders. Prevalence of oligomenorrhea in the Eastern zone, which is unfavorable environment. The levels of relative and absolute СD3+, СD4+ and СD8+ contents, the absolute amount of СD19+, FP, IgA, IgG, elevations of cortisol, testosterone, FSH, LH, and 17-OH-progesterone have been decreased. Indicators show the impact of the anthropogenic load on gynecological morbidity.

PREVALENCE OF ANTINUCLEAR ANTIBODY IN CHILDREN WITH REACTIVE ARTHRITIS

Abstract

Children with reactive arthritis (n=26) of 2-17 years old were examinated. Antinuclear antibodies were determined by indirect immunofluorescence assay, antibodies to extractable nuclear antigens were defined by ELISA. It was found that 12 patients (46 %) with reactive arthritis had diagnostic titers of antinuclear antibodies detected by indirect immunofluorescence assay. Homogeneous and speckled fluorescence patterns were observed in patients with reactive arthritis. The prevalence of antibodies to extractable nuclear antigens in children with reactive arthritis was low (8 %).

Abstract

Aim. To study the features of the system of hemostasis and the functional activity of neutrophils in patients with atherosclerosis of unpaired visceral branches of the abdominal aorta, depending on the presence of type 2 diabetes. Methods. The study included 87 patients. The first group included 32 patients with atherosclerosis of vessels of the plannonic pool and type 2 diabetes, to the second group of 55 patients without type 2 diabetes. Results. The median activity of Wf in the group of patients with type 2 diabetes was 193,5 %, significantly exceeding the parameters of the second group of patients 160,0 %, (p = 0,002). The phagocytic number of neutrophils of the group of patients with type 2 diabetes significantly exceeded the similar index of the second group of patients (p = 0,01). Рositive correlation was found between the activity of von Wf and the intensity of phagocytosis of peripheral blood neutrophils (r =0,723; p=0,04), and phagocytic number (r=,771, p=0,025). Conclusion. In patients with atherosclerosis of the visceral branches of the abdominal aorta and type 2 diabetes, higher values of the phagocytic number of peripheral blood neutrophils and Wf activity were revealed.

Abstract

It was made a model for studying in vivo induced immunosuppression, which often occurs with secondary immunodeficiencies. In the process of its creation, the activity of phagocytic blood cells was studied, as well as the characteristics of the cytokine profile. However, little is known about the biochemical parameters of peripheral blood. The conducted studies showed that when induced immunosuppression as a result of changes in the activity of mononuclear cells, the content of the total protein in the serum of peripheral blood increases.

Abstract

The specific features of differentiation of dendritic cells (DC) of the thymus in children with depending on the type of cyanosis at congenital heart diseases (CHD) have been studied. It has been established that the accumulation of DC in the medullar and cortical compartment of the thymus occurs at CHD, especially in the case of blue types of CHD. When comparing the blue and white types of CHD, a change in the content of myeloid and plasmacytoid DC of the thymus was revealed. Thus, hypoxia affects the differentiation of the DC at thymus at CHD.

Abstract

The most important criterion of implantable vascular prostheses biocompatibility is the intensity of local and systemic inflammatory reactions to the graft material. Purpose: To evaluate the intensity of local and systemic inflammatory reactions of the organism in the acute period after implantation of small diameter vascular prostheses created on the basis of different biodegradable polymers. Obtained by electrospinning biopolymer prosthesis were implanted in the abdominal aorta of rats: I - polycaprolactone (PCL); II - copolymer of polylactic acid/polycaprolactone (PLC 7015); III - copolymer of polylactic acid/ polycaprolactone with polyglycolide (PLC 7015-PGS); IV - polylactic acid/polycaprolactone (PL-18PCL); V - polycapro-lactone/polyglycolide (PCL-PGS); VI - sham-operated control group. Grafts explantation was performed on the 14th day. The intensity of the systemic inflammatory response was assessed by serum level of IL-6, IL-10 and MCP-1, the intensity of the local inflammatory reaction - by HE-stained grafts sections analyzing. Prosthesis implantation in all cases was accompanied by the local inflammatory reaction development that was most pronounced in the case of PL-18PCL and PCL-PGS. Elevated level of MCP-1 was observed only by PLC 7015 grafts implantation. All studied polymers could be used as a stock for small diameter vascular prostheses development.

CBA mice were immunized parenterally with the NIBRG-14 vaccine strain of A/H5N1 influenza virus, and after 2 weeks they were infected with a high dose of the live IDCDC-RG2 strain of the A/H5N1 influenza virus. The course of the infection was severe and accompanied by high mortality. The injection of antihistamines (a mixtures of H1 and H2 histamine receptors blockers), at the time of infection, resulted in a 10% reduction in mortality (p<0.05). Data indicate that the severe course of influenza infection in vaccinated animals at least partially depends on the mast cells and histamine they release presumably upon activation by IgG antibodies and the virus.

ASSESSMENT OF RESISTANCE OF PRO-BIOTIC STRAINS OF MICROORGANISMS TO ESSENTIAL AND TOXIFEROUS HEAVY METALS

Abstract

Data on the experimental studying of influence of salts of heavy metals on growth of probiotic strains of bacteria of the sort Bacillus are presented in article. During the made experiments it was established that presence of various salts of heavy metals differently influences dynamics of body height of the studied microorganisms. But despite it, the common regularity in influence of the same ion of metal on all studied microorganisms is observed. Presence of ions of lead and iron has the stimulating effect on body height of all studied microorganisms, and presence of ions of a cobalt and cadmium oppresses them body height. Presence of ions of Zincum and manganese does not exert impact on dynamics of growth of the studied strains, the exception makes only B. licheniformis for which Zincum is a growth factor.

IMMUNOREACTIVITY OF PATIENTS WITH CHRONIC VIRAL HEPATITIS B AND C WITH THE DEVELOPMENT OF SYSTEMIC AUTOIMMUNIZATION

Abstract

Screening studies among 232 patients with chronic viral hepatitis B and C revealed systemic (organ-specific) autoantibodies in 43.8 % and 30.2 % of cases, respectively. When studying the characteristics of immune response of patients it was established that in case of chronic viral hepatitis B pathogenetic bases of development of autoimmune disorders of a systemic nature are the stimulation of production of antibodies by B lymphocytes, and a decrease in elimination of the pathogen, with a weakly expressed antiviral response. In chronic viral hepatitis C, autoimmune disorders of a systemic nature develop against a background of a deep T-cell immunodeficiency state and an inadequate antiviral response. And also with systemic autoantigenemia, along with high indicators of circulating immune complexes, which indicate activation of antiviral protection, there is a deficiency of phagocytic activity of neutrophils, which further aggravates the immunopathological process.

Abstract

The article is devoted to the investigation of the correspondence of the genotype of the IL-28B gene to the virological, biochemical and immunological parameters of patients with chronic viral hepatitis C in the course of antiviral therapy of pegylated interferon α and ribavirin. It was found that with the genotype of the CC gene of the IL-28B gene at the locus rsl2979860, in which the non-genotype of the hepatitis C virus was most often detected, self-healing and high efficacy of PVT are most often observed. On the part of the organism’s reactogenicity, this was accompanied by a more pronounced cytolytic syndrome and a high immunological reactivity of the organism, manifested in an increase in the expression of cytotoxic lymphocytes CD8+, activation markers CD23+ and CD38+.

Aim: to assess clinical value of antibodies to Herpes Simplex Virus (HSV) in outpatients with head and back pain. Cross sectional study included 45 female outpatients aged from 40 to 60. Two or more HSVinfec-tion exacerbations per year were registered in 25 persons (Group1) while 20 women had no exacerbation during previous year (Group 2). General practitioner and neurologist examination have been performed accompanied by IEA IgG anti-HSV type 1,2 antibodies assessment. Results: Group 1 showed 5-fold higher IgG anti-HSV antibodies level and post-herpetic neuralgia frequency then Group 2. High IgG anti-HSV antibodies level was associated with increased odds of HSV infection exacerbation (OR = 1.8; 95 % СШ 1.2-2.6).

Abstract

Aim of the study: to investigate the effect of erythropoietin (EPO) on the parameters of cellular immunity in preterm infants (PI) of early preterm birth (EPB). The study was performed on 15 full-term infants and 25 PI from EPB (gestational age 28 weeks - 30 weeks and 6 days). EPO was applied 6 days of life at a dose of 200 IU/kg every 48 h. Number of subpopulations of leukocytes in blood was determined by flow cytometry with antibodies of system “CytoDiff”. It was found that PI from EPB at 8 and 18 days of life reduced the representation in blood of B-lymphocytes, T-helper cells, T - and NK-cells; on the 8th day of life increases the number of monocytes and their precursors decreases the number of eosinophils; 18 day - reduced the number of monocytes, mature neutrophils, increases the number of mast cells, precursors of B-and T-lymphocytes. The use of EPO leads on the 8 days of life to increase in blood the total number of lymphocytes due to T - and NK-cells, the number of immature granulocytes; 18 day - to increase the total number of lymphocytes at the expense of B-lymphocytes, T-helper cells, T - and NK-cells, increasing the number of mature neutrophils, immature granulocytes, the precursors of monocytes.

Abstract

The aim of this work is to investigate the effect of transdermal film (TDF) with erythropoietin (EPO) on the parameters of innate and adaptive immunity in experimental thermal injury (TI). The experiment was carried out on 70 white nonlinear Mature rats, which were divided into 3 groups: 1 (n=10) - intact control, 2 (n=30) -TI, 3 (n=30) -TI with the imposition of TDF with EPO. TI IIIA degree with an area of 3.5 % was modeling by immersion of the site interscapular region of the animal in clean water with a temperature of 98-99°С for 12 s. the Study was performed at 3rd, 8th and 14th days TI. The number of neutrophils in the blood was determined by hematological analyzer “Medonic - M 16” (Sweden). The absorption ability of neutrophils blood was examined using of latex particles, the oxygen-dependent metabolism in spontaneous and induced NST-test. With the help of a flow cytometer “Navios” in the blood determined the number of CD3+ and CD45RA+ lymphocytes. The death of lymphocytes was assessed by staining cells with fluorochrome conjugated with annexin V (Annexin-5-FITC) and 7-aminoantipyrine D (7-AAD). It was found that experimental TI in the blood increases the number of neutrophils, their phagocytic and NST-reducing ability, reduced number of CD3+ and CD45RA+ lymphocytes at 5th, 8th and 14th days, increases in blood lymphocyte count with evidence of apoptosis and/or necrosis at 3rd and 8th days. The use of EPO in the composition of TPF in the TI did not accompanied by a change in the number of neutrophils in the blood at 3rd, 8th and 14th days, leads to a decrease in phagocytosis activity at day 14th, the intensity of phagocytosis - on the 8th day, activity and the intensity of spontaneous NST-test at day 14th, the induced NST-test - 3rd, 8th and 14th days; increase of CD3+ lymphocytes in the blood on the 8th day, the reduction in the number of blood lymphocytes with signs of apoptosis and/or necrosis at 14th day.

Abstract

In this work results of a research of a functional condition of a peripheral leucocytal link of a blood at the students who are studying at the first and second courses of medical university and sectioned into 3 groups are presented. The first group was made by the students belonging to I and II groups of health and, studying according to the main program of physical training; the second group - athletes-volleyball players; the third group students made III and IV groups of health studying according to the medical physical culture program. At the end of the first year at students of the first group indicators of a leucocytal link didn’t differ from control sizes, and after the second year of training all indicators were enlarged in comparison with indicators of the first year of training. At students of the II group (athletes) the reliable augmentation of all indicators of a leukocyte series of a blood on the second year of training was recorded. In the third group on the second year of training, engaged according to the medical physical culture program, reliable differences of indicators of a leucocytal link of a blood in comparison with students of the main group, demonstrating to what testifies weren’t taped: occupations by physical culture promote conservation of normal functioning of immune system.

Abstract

It has been established that the level of IL-6 in patients with subcompensated CP (class B) was significantly higher than in the compensated stage (p <0.001), and also had significant differences in patients with subcompensated and decompensated CP (p <0.001). The concentrations of VEGF and G-CSF significantly increased at the stage of severe cirrhosis (p <0.001 and p = 0.04, respectively). Proinflammatory cytokine TNF-α and anti-inflammatory IL-4 significantly increased during transition to the subcompensated stage of the disease (р=0.01 and р=0.04, respectively). There are no reliable differences between allelic variations of polymorphic gene loci TNF (G4682A), IL-6 (C174G) and VEGFA (G-634C), associated with liver cirrhosis severity levels, detected by us.

In this study the analysis of the immune status of the elderly patients with acute cholecystitis was carried out before surgery and on the 7th day afterwards. A reduction in the number of T-lymphocytes was found before surgery. On the 7th day after surgery, the down-regulation of the T-cell immunity was observed that manifested by the twofold decrease in the populations of T-helpers and T-killers. The use of immunomodulatory drug roncoleukin during first days after surgery allowed to increase the number of T-lymphocytes and thus to prevent the progression of secondary immunodeficiency in the elderly patients.

Abstract

Growth and viability in different culture media after thawing of hybridoma BAP3 cells that produces monoclonal antibodies against pregnancy-specific b1-glycoprotein (PSG) were studied in vitro. We show that cultivation in [DMEM+12 % FCS] leads to maximal cell growth (1,8-fold in two days) with retention of viability of more than 70 %. There were 1,1-1,2-fold growth and viability of 55-59 % in [DCCM-1]-based media and 1,6-fold growth in two days in [RPMI-1640+12 % FCS]. Therefore, application of [DMEM+12 % FCS] medium seems to be the most appropriate for cultivation of hybridoma BAP3 cells after their thawing from cryopreservation.

Abstract

To assess the functional activity of monocytes in preterm infants of different gestational age (22-27.6 weeks and 28-31 weeks) in the dynamics of the neonatal period, cord and peripheral blood were examined. All children with an ENMT at birth had a decreased willingness of effector cells to participate in intercellular interactions (decrease in the number of CD14+ CB 282+ and CD14+ CD157+ monocytes), in infants of gestational age 22-27 weeks - to antigen presentation of the pathogen (decrease in HLA receptor expression -DR+). At the end of the neonatal period, the functional activity of monocytes is restored, the number of CD14+ HLA-DR+ cells exceeds those of full-term children.

Semaphorin 3A (Sema 3A) is known to be an axon guidance molecule and is implicated in the development of nervous system. The aim of the study was to evaluate the influence of Sema 3A on functions of murine thymocytes, such as: proliferation, migration and adhesion to thymic epithelial cells. Sema 3A suppressed thymocyte proliferation at 100 and 200 ng/ml, possessed a chemorepellent activity towards thymocytes in the transwell system, and inhibited thymocyte adhesion to murine thymic epithelial cell lines cTEC 1-2 and mTEC 3-10. Thus, Sema 3A is a negative regulator of important thymocyte functions and may essentially affect the process of thymocyte maturation.

MOLECULAR DIAGNOSTICS OF IGE REACTIVITY TO BIRCH POLLEN ALLERGENS IN CHILDREN WITH ALLERGIC RHINITIS

Abstract

The aim of this study was to detect sIgE to isoallergens of birch pollen and to evaluate a contribution of some homologous food allergens to the development of sensibility in AR patients. There were 21 blood sera of AR patients and 20 blood sera of healthy persons without allergy symptoms. The total IgE levels and sIgE to allergens were measured by the ImmunoCAP method (Phadia, Sweden), as well as by the ELISA kits (Alkorbio, Russia). We detected high total IgE levels, sIgE to allergens of birch pollen, apple, celery, as well as to recombinant allergens of birch Bet v 1, Bet v 2 and soybean - Gly m 4 in AR patients. Correlation analysis showed the direct valid dependence between the sIgE levels to birch isoallergen Bet v 1 and soy isoallergen Gly m 4 (r=0,84; p<0,05). There was also the direct valid correlation between the sIgE levels to birch isoallergen Bet v 1 and allergens of apple and celery (r=0,67 and 0,57, accordingly). Our finding permits assert that cross-reactivity can be important in exacerbation of clinical symptoms in AR patients including progress of oral allergy syndrome.

SPECIFICITY OF PERIPHERAL BLOOD PARAMETERS DEPENDING ON THE LEVEL OF SPORTS QUALIFICATION OF SPORTSMEN

Abstract

The investigation is aimed to problems of dynamic changes associated with a progress in sports qualification of sports professionals, and changes in peripheral red blood cell-associated parameters, probability corresponding to positive or negative signs of blood system adaptation to aerobic or anaerobic physical activity conditions. The optimal level of red blood cell-associated parameters, that meets the elevated oxygen tissue consumption demands, was observed in groups of Masters of sports and candidates for Master of sports. In groups of wrestlers and boxers some variations in red blood cell-associated parameters were also observed, with mostly lack of statistically significant differences between sports, sport qualification, or other parameters investigated, indicating no or little impact of sports specificity factors on red blood cell-associated parameters. The values of “leucocytes blood formula entropy” in groups of skiers, swimmers and wrestlers were within normal range, thus indicating a relatively adequate state of adaptation of blood system and it’s regulation in response to increased physical activity, but in boxers group they were out of normal range, probably due to negative side effect of adaptation processes or even a pre-morbid (pre-disease) state. The values of “integral coefficient of blood deterioration” also indicated on worse changes, being statistically significant in group of boxers only, thus reflecting deviations of functional activity, probably due to negative adaptive reaction accumulation as a result of specific factors in this sport. Conclusions: Progress in sports qualification mostly of aerobic (cross-country skiers, swimmers) or anaerobic (wrestlers, boxers) physical activity, was accompanied with changes in red blood cell-associated parameters, thus with sufficient probability corresponding to specific effects of aerobic or anaerobic physical activity on sportsmen.

Abstract

Recently, we detected a rheumatoid factor, called regulatory, whose production prevents development and causes a remission of experimentally-induced autoimmune diseases. The aim of this study was to elucidate the mechanism of induction of antigenic determinants for regulatory rheumatoid factor onIgG and Fc fragments of human IgG. It was found out that antigenic determinants for regulatory rheumatoid factor are formed in the hinge region of IgG uponthe reduction of sulfhydryl bondby treatment of the IgG molecule with polyethylene glycol or dithiothreitol. In response to the immunization of rats with native human IgG, the production of antibodies similar in specificity to human regulatory rheumatoid factor occurs, suggests that there is an endogenous mechanism for the formation of neoantigenic determinants for regulatory rheumatoid factor

Abstract

Clinical experience showed that use of the probiotic medicine “Sporobakterin” promotes not only the fissile elimination of ions of lead from an organism to 58,9 % (from an integument to 67,9 % muscular tissue to 47,1 % and a bone tissue to 61,5 %), but also prevents development of degenerative changes of internals that in our opinion has direct dependence. On by it application of Sporobakterin at treatment of intoxication lead interferes with development of inflammatory process in an organism what morphological, hematological and biochemical indexes of blood of the experimental animals testify to, and also have the stimulating effect of factors of nonspecific resistance of an organism.

DISTRIBUTION OF TWO-LOCUS HAPLOTYPES OF HLA-DRB1-TNFA-308 IN PATIENTS WITH ULCERATIVE COLITIS OF THE RUSSIAN POPULATION OF CHELYABINSK REGION

Abstract

The analysis of the distribution of HLA-DRB1-TNFA-308 two-locus haplotypes frequencies in patients with ulcerative colitis (UC) of the Russian population of the Chelyabinsk region is carried out in this article. The 19 haplotypes were identified, among which 4 haplotypes, containing -308*G TNFA allele, were characterized by high incidence rates in groups of UC patients and healthy individuals. A peculiarity of the UC patient group was the presence of two haplotypes containing the rare allele with the substitution -308*A TNFA: HLA DRB1* 01-TNFA-308*A; HLA DRB1* 13 -TNFA-308*A. These haplotypes were not found in the comparison group of healthy individuals. These haplotypes can be considered as markers of susceptibility to ulcerative colitis in the Russian Chelyabinsk region.

A new mechanism for activating the differentiation of naive T cells in Th17 in the presentation of autoantigens with B cells in multiple sclerosis has been suggested.

Abstract

Human papillomavirus infection (HPV infection) is equally widespread among women and among men. The purpose of this study was to determine the levels of main immunological parameters of the secretory immune system in women and men from 38 couples infected with HPV of high oncogenic risk, with different types of viral load. The object of the study in women was cervical mucus, men - ejaculate. Results. Both women and men from couples with HPV each time a viral load was determined by the increase in the number of IL-2 and IL-8, decrease of IL-4 and TNF-α. When significant concentrations of HPV DNA in women they have increased the number of sIgA, men elevated were IL-φ, IFN-α and IFN-γ.

The effect of methylprednisolone (MP) on the functional activity of CD4+ CD45RO+ T cells obtained in patients with rheumatoid arthritis (RA) and healthy donors in the in vitro system was studied. It has been shown that MP significantly reduces the number of TCR-activated СD4+СD45RO+СD25+ and СD4+СD45RO+СD71+ T-lymphocytes in cultures of RA patients and in healthy donors. MP, reducing the number of СD4+СD28+ T cells, promotes the formation of terminal-differentiated memory effector T-cells.

Abstract

In pediatric patients, pneumonia is one of the most urgent problems of modern medicine and the most important single cause of death in children worldwide. One of the leading causes of pneumonia at the present time is the change in the immunological reactivity of the organism. To identify features of the immune status, namely cellular and humoral links of immunity in children, we examined a total of 86 children aged 1 year to 18 years. All the children were divided into 2 groups: the first group of 60 children with a diagnosis of community-acquired pneumonia, the second group consisted of 26 healthy children of corresponding age and sex. Thus revealed the imbalance of cellular and humoral immunity in children with community-acquired pneumonia. Studies of the immune status in patients with pneumonia indicate a decrease in levels of serum immunoglobulins and phagocytosis, increased levels of circulating immune complexes.

Our results confirmed that the screening of cytokines prior to transplantation may be used to predict the subsequent development of complications.

Abstract

Chemotaxis and other factors of innate immunity are involved in metastasis (such as TLRs), it is advisable to study the effect of both TLRs and chemokines on the chemotaxis of tumor cells. The aim. Study and compare the chemotaxis of mononuclear cells and tumor cells towards TLRs ligands and CXCL12. Materials and methods. Chemoattractants were synthetic TLR ligands (DNA_lig, RNA_lig) and CXCL12. The study of cell migration was carried out in dynamics with the help of the Boyden chamber. Results. ^emotaxis of MNC from healthy donors exceeds the chemotaxis of tumor cells by 4 times and significantly differs by 1,6 times from spontaneous migration. Directional chemotaxis of MNCs of healthy donors in 1,7 and 1,5 times greater than the migration of MNCs of healthy donors after 60 minutes and 24 hours, respectively, relative to migration to DNA_lig and RNA_lig. Conclusion. From the results obtained, it can be seen that CXCL12-induced chemotaxis is higher in MNCs of healthy donors than in tumor cells. Similarly, chemotaxis relative to DNA_lig and RNA_lig is significantly lower than in the control. Thus, it can be said that TLRs ligands may inhibit migratory activity.

The levels of serum cytokines were investigated in 17 patients with aseptic necrosis of the femoral head before surgery total joint replacement aged between 33 and 57. A significant increase in the blood compared to normal levels of TNF-α, IL-17, IL - 4, IL-10 as well as the decrease of IFN-γ was determined. The identified imbalance of cytokines which are positively and negatively regulating the osteoclastogenesis may contribute to an increased resorption of bone tissue in this pathology.

Abstract

With the purpose of revealing the features of the innate immunity of premature infants with severe hypoxic-ischemic injury of the central nervous system, depending on the gestational age, a survey of deeply preterm infants born at the time of very early and early premature births was conducted. It was established that in all premature infants with severe hypoxic-ischemic CNS lesions, the level of expression of adhesion molecules on monocytes (CD14+CD11b+) and an increase in the level of IFN-γ and IL-8 at birth was observed at birth, regardless of the gestational age. The peculiarity of the immune response of children born in the term of early premature births is a decrease in the relative number of activated monocytes of CD14+CD64+ and CD14+HLA-DR, which indicates a disruption in the relationship between the cells of congenital and adaptive immunity and may be the cause of bacterial complications and a prolonged course of inflammatory Process.

Abstract

Acute cold stress (4°C, 4 hours) led to inhibition of the production of ROS (reactive oxygen species) in stimulated cultures of peritoneal macrophages of the mouse. The effect of acute cold stress was not modified by the administration of naloxone. Chronic cold stress (4°C, 4 hours, 7 days) led to stimulation of ROS production by peritoneal macrophages of mice in the presence of zymosan. This effect is partially canceled by the administration of naloxone.

Abstract

The aim of this study was to study the effect of the injectable form of platelet rich autoplasma on immunological indices of the oral cavity to fixed prostheses patients. The number and viability of leukocytes and phagocytic function of neutrophils of oral fluid to fixed prostheses patients was significantly lower than to patients without dentures. However, after the injection of platelet rich autoplasma indicators of cellular immunity to patients with fixed prostheses approached that of patients without dentures, which indicates a positive immunological effects of injection of platelet rich autoplasma.

It was established that the dosage forms prepared with the use of polyethylene glycols provided a uniform plank mass. Taking into account the process of liberation in the conditions of dissolution temperature as well as spectrophotometric determination, the technology of the novel form of the drug form - suppositories with interferon alpha in combination with antioxidants.
